RESUMO:

Brazil is the largest producer of sugarcane in the world, yet there are many production losses that can be caused by both environmental factors and entomological factors. The pest of Telchin licus licus (giant borer) has a higher occurrence in the Northeast region and its control is quite difficult, since the larva closes the hole just after the cane cut, which makes the chemical control inefficient. Manual harvesting is the only efficient method, but labor costs are very high. The use of Bacillus thuringiensis as an alternative to chemical control has been shown to be quite efficient, being the most used microorganism in insect control. In view of the above, this research project aims to isolate B. thuringiensis bacteria from the sugar cane plant, root, central cylinder and giant drill larvae liquid and to study the entomopathogenic capacity of these bacteria against larvae of the giant drill. To identify the species of Bacillus will be done Gram staining, evaluation of spore production by the isolates. A study of Bacillus bacterial diversity present in sugarcane plants and in the intestine of the giant borer will be performed using the REP-PCR technique. Bacterial isolates will be taxonomically characterized by amplification and sequencing of the 16S RNAr region. Isolates that show sequences similar to B. thuringiensis sequences deposited in the database will have the DNA amplified using primers specific for cry gene amplification. An evaluation of the bacteria's ability to colonize sugarcane tissues will also be carried out. In addition, in vitro tests will be carried out, where the larvae will be fed an artificial diet inoculated with the bacteria, in vivo test, in which the larvae will be fed with stems inoculated with the bacteria and trials in sugarcane producing areas affected with the pest. At the end of these tests, the mortality rate of the larvae will be evaluated and, with the results obtained, the most promising strains will be selected to be used in the control of the giant burrow in the greenhouse and field.
