Brazil stands out as a major world producer of the sugarcane culture, in addition to its use in the food industry, it is used for the production of ethanol, its importance stands out in the energy scenario due to changes in Brazilian legislation through the Law No. 10,203 of 2001, which establishes the mandatory use of the addition of anhydrous ethyl alcohol to gasoline. In high-yield agriculture, considerable applications of mineral fertilizers are necessary to maintain the productivity of agricultural crops. Currently, Brazil is considered the fifth country that most uses fertilizers to increase crop productivity. Scientific researches reveal that the excessive application of fertilizers can cause eutrophication of water bodies, producing environmental damage. In this sense, the knowledge of the interaction between plants and bacteria can mean advances for the development of inoculants for the cultivation of sugarcane, reducing the use of chemical inputs. The endophytic bacteria Gluconacetobacter diazotrophicus PAL5, was initially isolated, in Brazil, from sugarcane stalks. Several studies highlight the use of G. diazotrophicus as a potential use in several agricultural crops. In this thesis, there is a quantitative proteomic analysis for the identification of differentially expressed proteins of nitrogen fixing bacteria G. diazotrophicus strain PAL5, when grown in LGI-P medium modified with 0.5% glucose, supplemented or not with 50% liquid of sugarcane apoplast variety RB867515 (responsive to inoculation with bacteria from sugarcane inoculant). The total proteome resulted in 1156 proteins for each treatment, while statistical analysis analyzed 58 proteins differentially expressed between each interaction. Statistical analyses based on Vulcano plot indicate that there is a significant difference between the level of expression of regulated proteins, with the analysis of Clusters of Orthologous Groups (COG), it was found that compared to the conditions of growth of the bacterium, the medium composed of liquid apoplastic alters expression by suppressing proteins related to cell, membrane, and cell envelope biogenesis, and overexpresses proteins related to post-translational modification, protein recycling and chaperones. According to scientific descriptions of the role of the apoplast in the regulation of endophytic bacteria, it is observed that the intercellular liquid of sugarcane stalks alters the expression of proteins of G. diazotrophicus strain PAL5, relevant between the plant-bacterial interaction.
