SUMMARY:

The target species of this project is the vinca (Catharanthusroseus (L.) G. Don), belonging to the Apocinaceae family. Its probable origin is the island of Madagascar. The great importance of vinca lies in the production of alkaloids such as vincristine and vinblastine, widely used in the treatment of lymphomas, leukemias, breast, lung, testicular and Kaposi's sarcoma. The fact that some of the alkaloids are not synthesized in the laboratory underscores the importance of cultivating vinca in the field. However, the extraction of alkaloids from vinca plants has as a barrier the high production costs due to their low yield in the culture. Therefore, genetic improvement programs for C. roseus are an important strategy for the selection of superior genotypes for the production of different alkaloids in the species. The general objective of this thesis project is to know the behavior of vinca genotypes regarding their stability and biomass production potential during two consecutive years (Phase 1). The superior genotypes selected in Phase 1 will be studied for vincristine alkaloid concentration (Phase 2). For this purpose, 40 genotypes listed as superior in biomass production will be studied, conducted in a greenhouse. In the first stage, 19 morphoagronomic variables related to plant size, leaves and biomass production will be measured. At this stage, seven variables related to secretory cell structures will also be measured from paradermal histological sections that will be submitted to histochemical test for alkaloid detection with Dragendorff. Measurements will take place every 120 days for two consecutive years. The statistical analyzes of Phase 1 will be as follows: study of the direct and indirect effects through Spearman's Correlation and Path Analysis, obtaining applied regression models in order to know the general and individual behavior of the genotypes in relation to a given variable, estimation of genetic parameters and variance components via REML/Blup and interpretation complementary to Blup using Blup/PCA methodology. The genotypes selected as superior in this step will proceed to Phase 2. In this step, HPLC analysis will be performed to quantify the vincristine alkaloid. The genotypes that stand out as superior in terms of the production of the referred alkaloid will be submitted to genotyping to estimate the genetic distances between them. It is expected at the end of this work to point out superior genotypes for the characters related to biomass and vincristine alkaloid production. With this information in hand, proceed with the Crop Genetic Improvement Program developed in the Department of Plant Science at the Institute of Agronomy at UFRRJ.
