Gerbera is one of the most important cut flowers sold in the world, occupying fifth place among flowers in this rancking. The modern gerbera cultivars show great variability due to the genetic breeding, which began with crossing of two South African species, Gerbera jamesonii and Gerbera viridifolia, originating Gerbera hybrida. Micropropagation is the most widely used propagation method commercially for this species, because produces large numbers of seedlings faster than natural one. The objective this research was to establish a protocol for gerbera micropropagation with purpose providing subsidies for the conservation and multiplication genotypes, to analyze in vitro culture for mass seedlings production, as well to evaluate your genetic diversity. For this purpose, genetic diversity of 14 gerbera germplasm was evaluated based on morphological leaves and flowers descriptors. Micropropagation protocol of the genotypes was also carried out, with two experiments. In the first was developed a protocol comprising all phases micropropagation, since: in vitro establishment used leaves with petiole as explant; in multiplication, different concentrations BAP (6-benzylaminopurine) and ANA (naphthalene acetic acid) were tested; in vitro rooting different ANA levels, sucrose concentrations and MS culture medium were tested, as well genotypes rooting in the MS and B&G® culture medium and acclimatization were analyzed. In the second micropropagation experiment, cytokinin/auxin hormone balance of the genotypes was optimized through different BAP and ANA concentrations. Considering results, observed formation of several groups gerbera genotypes when tested at Euclidian and Mahalanobis distance for quantitative data, and Gower algorithm was the most efficient because provided formation contrasting groups for qualitative and quantitative characteristics together analyzed. Principal component analysis identified that quantitative descriptors referring to the width ligated flower external radius, leaf width, stalk length, sheath height and disc diameter were least contributors to genetic diversity. During micropropagation protocol, seven genotypes were lost due to fungal and bacterial contamination and diferences between genotypes were observed for the number of shoots produced. On rooting was verified that complete MS medium, supplemented with 30 g L-1 of sucrose and without ANA provided to obtain bigger and better rooted seedlings. Was also verified that B&G® medium provided better responses for rooting and seedling size and during acclimatization the survival rate was high. Finally, the evaluation hormonal balance indicated the best BAP and ANA concentration for each gerbera genotype referring to the variables number of shoots per explant and average length of shoots. In this context, was verified existence genetic diversity gerberas collection of the UFRRJ and established an efficient protocol gerbera mass production.
