SUMMARY:

Antimicrobial resistance (AMR) is a critical global public health challenge that requires an integrated approach grounded in the 'One Health' concept, which acknowledges the interdependence of human, animal, and environmental health. In this context, although soil harbors a highly diverse microbial community and a natural resistome potentially transferable to both commensal and pathogenic bacteria, it remains frequently overlooked in AMR surveillance efforts. This study aimed to evaluate the structure of bacterial communities and the presence of antimicrobial-resistant bacteria in soils with varying land-use histories, including agricultural areas and secondary Atlantic Forest fragments in São José do Vale do Rio Preto, RJ, Brazil. Total soil DNA was extracted and subjected to 16S rRNA gene sequencing, while bacterial isolation was performed using selective media containing various antimicrobials. Agricultural management altered the structure of bacterial communities and influenced the profile of genera isolated on antimicrobial-containing media. Seven of the ten tested antimicrobials predominantly selected for bacterial genera with predicted susceptibility phenotypes—particularly in response to ciprofloxacin, gentamicin, and sulfamethoxazole-trimethoprim. These findings underscore the role of agricultural environments as significant reservoirs of resistance genes with the potential to disseminate to human pathogens and highlight the urgent need to include managed soils in AMR monitoring and mitigation strategies.
