RESUMO:

The Artificial Lake Açu located on the campus of UFRRJ is a space for students, staff and visitors that is bordered by the Agronomy, Biology and Veterinary Institutes, as well as the Rector's House (UFFRJ, 2013). This site presents a great diversity of plant and animal species such as capybaras (Hydrochoerus hydrochaeris Linnaeus, 1766) (Rocha et al., 2017), as well as being a propitious place for reproduction and development of parasites such as ticks. The star tick, Amblyoma sculptum Berlese 1888 (Nava et al., 2014), is the host of Gram negative Rickettsia rickettsi, which causes fever, a disease that is lethal to humans and is the main parasite of capybaras (Szabó et al., 2013). The objective of the research project is the hemolymph analysis of infected and non-infected Rickettsia spp. Ticks, collected at the campus of UFRRJ, located in the municipality of Seropédica, in search of chemical markers that indicate the contamination of ticks by the microbe. The ticks will be collected from the surroundings of Lake Açu, through traps of dry ice and identified according to Barros-Battesti et al. (2006) and Nava et al. (2014). Specimens will be kept in greenhouses B.O.D. (32ºC per 24 hours) for the multiplication of Riquettsias for the extraction of hemolymph from adults of A.sculptum Angelo et al. (2010) and afterwards the following procedures will be performed: detection of the presence of microorganisms compatible with Rickettsia Burgdorfer (1970), visualization of Giménez bacteria (1964) and confirmation of the presence of pathogenic Rickettsiae for fever through DNA extraction and PCR. For the investigation of chemical markers in hemolymph, the hemolymph samples will be subjected to extraction, partition, derivatization and GC-MS analysis according to Roessner et al. (2000) and Lisec et al. (2006) with modifications. The investigation of the chemical profile of the hemolymph of ticks infected by the bacterium Rickettsia spp. Through chemical markers allows a mapping of the risk of contamination in a given locality, serving as useful information for public policies to combat spotted fever.
