RESUMO:

The chemistry of natural products, as well as other areas of chemistry, has sought to perfect its analytical methodologies with new techniques and equipment. The main considerations in this regard are to achieve results in less time, less costs of the analyzes with better results and from small amounts of matrix. The technique of dereplication has been used to identify special metabolites in complex matrices starting from very low mass. This technique relies on ultra- or high-efficiency chromatography equipment coupled to detectors such as ultraviolet, mass spectrometry and/or NMR. In the area of Chemistry of Natural Products, it has turned out to be a quick procedure to identify already known metabolites as well as to track new compounds. The identification of new compounds will certainly require other techniques of isolation and identification by 1H and 13C NMR, CD, and mass spectra analysis, for example. The objective of this project is the application of these techniques to the study of two species of Verbenaceae, Lippia alba and L. triplinervis, to identify the special metabolites. The Verbenacea comprise 90 genera which together total 2614 species, distributed throughout the world, in Brazil there are 47 genera, in which some stand out for their medicinal potential indicated by the popular wisdom and some of them already proven scientifically. The literature indicates the genus Lippia as a bioproducer of essential oils and fixed substances such as flavonoids, tannins, naphthoquinones, iridoids, saponins and terpenes. The genus Lippia stands out as one of the most expressive of the family, in which there are two specimens recognized by the Brazilian Pharmacopoeia as medicinal plants, Lippia alba (erva-cidreira or false Melissa) and Lippia sidoides (alecrim pimenta). The herbal from of the Brazilian Pharmacopoeia recognizes its anti-inflammatory, soft sedative, antimicrobial properties of the oral cavity, etc., of the Lippia species. Investigations already published justify these activities by the population besides volatile compounds, however it is known that with the methods used by the population besides volatile compounds there are other active constituents. By means of the above, we intend to evaluate qualitatively the compounds in the extract obtained by two popular techniques, tincture and infusion, varying the condition of the source material in fresh and dry leaves, keeping the stages of extractive processes controlled. The mapping of the phytochemical profile of the different obtained extracts will be through the deregulation, as mentioned above, with sensitive equipment such as GC-MS, GC-FID, UPLC-MS/MS or HPLC-UV. The identification of the metabolites will be done through the information obtained from the detection instruments hyphenated to the chromatography in comparison to the database produced through the literature information for the genus and the family, and interpretation of the data obtained by 1H and 13C NMR, uni and two-dimensional.
