SUMMARY:

With the aim of optimizing the ejaculatory function in animals that are unable to mount due to some type of illness, pharmacological ejaculation induction has been used for some years as an alternative method. The semen of domestic animals can be contaminated by microorganisms, however, there are few studies on the microbial flora present in the semen of breeders. The main negative effect that these microorganisms cause on semen quality is on sperm motility parameters, in addition to compromising acrosomal function and ultrastructural damage. Therefore, this work aims to evaluate the effectiveness of detomidine as an ejaculation inducer at doses of 0.10 μg/kg, 0.13 μg/kg, 0.16 μg/kg, in addition to comparing the microbiota of semen collected with this inducer and with artificial vagina. Four Mangalarga Marchador stallions will be used, with an average weight of 400 kg, age ranging from 3 to 15 years, all with no history of infertility. The stallions will be divided into four groups, with four semen collections every 48 hours per animal, with no ejaculation in the control group, the collection will be performed using an artificial vagina. All stallions will pass through all groups in the crossover system. Being distributed as follows: GI (n = 12) control group will receive 1.0 ml of saline solution, GII (n = 12) will receive 0.10 μg/kg of 1% detomidine hydrochloride (Detomidin®), GIII ( n = 12) will receive 0.20 μg/kg of 1% detomidine hydrochloride (Detomidin®), GIV (n = 12) will receive 0.30 μg/kg of 1% detomidine hydrochloride (Detomidin®). The ejaculate will be collected in a disposable plastic cone attached to a collection cup, held by an assistant close to the animal's foreskin, which will be performed within 1 hour. Subsequently, the semen will be submitted to motility, vigor, concentration, volume and microbiological analysis. The experimental design used will be the Latin square, with a repetition after ten days. In this design, the blocks are organized in two different ways, some constituting “rows” and others “columns” (Table 1). The statistical test used will be the Chi-square test (X2) with a significance level of 5%. The results of p ≤ 0.05 are considered to have statistically significant differences and p ≥ 0.05 are considered to have no statistically significant differences.
