SUMMARY:

Plants used in medicinal treatment for healing skin wounds and treating inflammation have often been applied without any scientific evidence of efficacy and with little knowledge about the molecular mechanism of the active compounds responsible for the effects. Wound healing is a complex process involving a series of events, such as different cell types (monocytes, endothelial cells, fibroblasts, and keratinocytes), cytokines (interleukins, TNF-α), growth factors, extracellular matrix, and cellular processes. These phases include homeostasis, inflammation, cell migration, protein synthesis, contraction, and wound remodeling. Therefore, the objective of this work is to evaluate the healing, antinociceptive, and anti-inflammatory activities of the latex and ethanolic extract of Hancornia speciosa Gomes. Many plant species are traditionally used in Brazil to improve skin wounds due to their anti-inflammatory properties. Hancornia speciosa Gomes is a species discovered in the Brazilian Cerrado biome, popularly called "mangaba" or "mangabeira". Its leaves are traditionally used for their antihypertensive and antidiabetic properties. This species is also traditionally used to treat fungal infections, tuberculosis, ulcers, and to stimulate liver function. The objectives of this work are to evaluate the antinociceptive action of the latex and ethanolic extract of Hancornia speciosa under conditions of acute pain; to evaluate the anti-inflammatory activity of the latex and ethanolic extract of Hancornia speciosa under conditions of acute inflammation; to elucidate the mechanisms responsible for the antinociceptive and anti-inflammatory activities of the latex and ethanolic extract of Hancornia speciosa; to evaluate the possibility of interference of motor performance on the antinociceptive effect demonstrated by the latex and ethanolic extract of Hancornia speciosa; To evaluate the characteristics of the healing process through analysis of scar retraction, histopathological analysis, and cytokine quantification under topical administration of latex and ethanolic extract of Hancornia speciosa, and to evaluate the acute toxicity of latex and ethanolic extract of Hancornia speciosa in mice and dogs.
