SUMMARY:

Plants used in medicinal treatment for healing skin wounds and treating inflammation have often been applied without any scientific evidence of effectiveness and with little knowledge about the molecular mechanism of the active compounds responsible for the effects. Wound healing is a complex process that involves a series of events, such as different cell types (monocytes, endothelial cells, fibroblasts and keratinocytes), cytokines, growth factors, extracellular matrix and cellular processes. These phases include homeostasis, inflammation, cell migration, protein synthesis, wound contraction and remodeling. Therefore, it is extremely important to discover new drugs that induce fewer adverse events, are more effective and facilitate patient adherence to treatment, for the success of the therapy. For these reasons, this work aims to obtain a product administered topically that can be used in the treatment of wounds with a view to reducing pain and improving the wound healing process. Many plant species are traditionally used in Brazil to improve skin wounds due to their anti-inflammatory properties. Hancornia speciosa Gomes is a species discovered in the Brazilian cerrado biome, popularly called “mangaba” or “mangabeira”. Its leaves are traditionally used for their antihypertensive and antidiabetic properties. This species is also traditionally used to treat fungal infections, tuberculosis, ulcers and to stimulate liver function. The objectives of this work are to evaluate the antinociceptive action under acute pain conditions; evaluate anti-inflammatory activity under conditions of acute inflammation; evaluate the characteristics of the healing process through the analysis of scar retraction, histopathological analysis and quantification of cytokines under the cutaneous administration of pure latex and ointments with increasing concentrations of latex and Hancornia speciosa leaf extracts in mice and dogs. The present project involves the use of scar retraction assessment models and in vivo histological analysis; in vitro models of cytokine quantification in mice. In dogs, antinociceptive and macroscopic evaluation of wounds in animals undergoing mastectomy.
