RESUMO:

Currently, the main objective of research related to the treatment of chronic pain and inflammatory conditions, conditions related to a high degree of morbidity and mortality in the veterinary clinical routine, has been the development of new substances with greater efficacy, lower toxicity and economic cost than the drugs currently available, and the compounds extracted from medicinal plants, especially aromatic plants, have aroused notable interest of researchers, regarding their chemical aspects and potential pharmacological properties. The present work aims to (1) confirm and quantify the possible antinociceptive and anti-inflammatory effects of nucleoside derivatives from monoterpenes derived from essential oils of Geranium, Bergamot and Citronella in ex vivo and in vivo models in mice of the Swiss line; (2) propose a structure-activity relationship based on the results obtained, and; (3) to assess the oral toxicity of the substances in order to propose the use of these substances in future clinical trials on pets. In a first phase, a three-step study will be carried out to establish and compare the ED50 of the three substances through the abdominal contortion for acetic acid and formalin tests (screening) to determine the most promising therapeutic potential compound. In a second phase, the following experimental models will be applied: tail flick; paw edema; Rotarod; Air pouch with quantification of cytokines (IL-1β, IL-6 and TNF-α) and total leukocytes; analysis of the activity of cycloxigenase 1 and 2 (COX-1 and COX-2) enzymes and in vivo toxicological evaluation by the model proposed by Lorke (1983). The statistical significance between the groups will be determined by the analysis of variance (ANOVA in a single pathway) followed by the Bonferroni test, assuming p values less than 0.05; 0.01 and 0.001 as significant. Thus, we believe that the study may contribute to the development of veterinary pharmacology and therapeutics.
