Universidade Federal Rural do Rio de Janeiro, Seropédica, RJ, 2021.This research aims to define the profile of microorganisms present in the feces of weaned foals; description and evaluation of the diversity of microorganisms in the post-weaning foals; identify a microbiota using next-generation sequencing; assess whether there are differences in the diversity of fecal microorganisms from horses and mules; and evaluate the effects of forage types on the foals' fecal microbiota. The research was conducted in a completely randomized design in a 2x3x2 factorial arrangement with the 1st factor - Equus caballus foals and the hybrid mule foals Equus caballus x Equus asinus; the 2nd factor - pastures with the cultivars Coast Cross, Florakirk and Tifton 85; 3rd factor - periods I (first period) and II (second period). Twelve freshly weaned foals were used, being six Mangalarga Machador horses and six donkey Pêga mules and crossbred mares. The foals were kept in paddocks, under rotational management in Cynodon dactylon pastures: Coast Cross, Florakirk and Tifton 85, being rotated to a new paddock every 20 days for four months. Six collections of fecal samples from animals and two collections from pastures were carried out at the end of each experimental period for chemical analysis. Microorganism-dependent techniques were used to obtain the counts of bacteria with fibrolytic functions. Total DNA extractions were performed from fecal samples and Illumina Miseq sequencing was performed. Bioinformatics analyzes were carried out with the raw data from the sequencing and the relative abundances of the different taxonomic levels, the Chao 1 richness and Shannon and Simpson diversity indices were obtained, the distribution of OTUs was evaluated and the treatment groups were compared. through Lefse. The performance variables of growth and body weight gain, bacterial counts of Streptococcus, Lactobacillus, total anaerobic and cellulolytic bacteria and relative abundances of different taxonomic levels were submitted to analysis of variance. Equine and mule foals showed differences in relative abundances at different taxonomic levels, in the number of OTUs and Shannon and Simpson diversity indices, in microbiological counts of Streptococcus and Lactobacillus and in the pH of fecal samples. Pastures of the genus Cynodon also provided differences in relative abundance, number of OTUs and Chao 1 richness index, total anaerobic counts, fecal pH, and measures of growth performance of foals
